将(N-吡咯)己基硫醇自组装在金膜上,并采用化学氧化聚合法使吡咯与羧基功能化吡咯衍生物在自组装薄膜上进行共聚,然后对共聚物进行羧基活化处理.探针DNA通过与共聚物膜之间的共价键作用而固定在其表面上,接着与靶向DNA杂交.同时采用傅立叶红外变换光谱仪和X-射线光电子能谱仪对共聚物在DNA固定前后的化学组成进行详细表征.采用电化学工作站对DNA在共聚物薄膜上的固定/杂交前后所引起的电化学性质变化进行测试.还采用等离子表面谐振仪在位考察了DNA在共聚物膜上的固定动力学过程,进一步探明DNA的固定行为.结果表明DNA在导电吡咯共聚物薄膜上的固定是一缓慢过程,同时DNA固定/杂交也会引起共聚物薄膜的电化学活性和电荷转移能力的降低.
Pyrrole (Py) and pyrrole derivative with carboxyl groups
1\|(2\|carboxyethyl)pyrrole (PyCA)
were copolymerized with 6-(1H-pyrrol-1-yl)hexane-l-thiol(PHT) self-assembled on Au surface by chemical polymerization to form a layer of the copolymer film
(poly(PHT-co-Py-co-PyCA)).The carboxyl groups of (poly(PHT-co-Py-co-PyCA)) were activated using N-(3-dimethylaminopropyl)-N-ethylcarbodilimide (EDC) and N-hydroxysuccinimide (NHS) and transferred to amide bonds by reacting with the amino groups in the amino-labeled probe DNA.In consequence
amino-labeled DNA could be immobilized onto the copolymer.Afterwards
the complementary target DNA can be hybridized with probe DNA.The chemical structure and components of poly(PHT-co-Py-coPyCA) before and after DNA immobilization were investigated by Fourier-transform infrared (FTIR) spectroscopy and X-ray photoelectron spectroscopy (XPS).The electrochemistry properties and the variation of the charges of copolymer film before and after DNA anchor were characterized by cyclic voltammetry and electrochemical impedance spectra.To detect the immobilization behavior of DNA
surface plasma resonance was used.The result revealed that it is a slow process for DNA anchoring onto the copolymer matrix and the electrochemical activity and the charges transfer decreased after DNA immobilization/hybridization.Furthermore
there is no non-specific adsorption between target DNA and copolymer.It could supply the theoretical and experimental base for the application of copolypyrrole film functionalized with carboxyl groups as DNA sensors.
